1.LGR4 and ZNRF3: Basic structure which can interact with all 4 kinds of RSPOs. 2.Linker: To improve the flexibility of RTAC. 3.FZD5 signaling peptide: To improve the secretion capability. 4.Human IgGH3 Fc: To improve the stability.

ZNRF3 and LGR4 are both extracellular domains of two membrane proteins that have been reported to interact with RSPO and connect them with a linker with optimized length.
